Omae wa mou shindeiru: meaning and grammar of the meme

"Omae wa mou shindeiru" means "You are already dead." Kenshiro says it in Fist of the North Star (Hokuto no Ken) after striking an opponent’s pressure points. His opponent hasn’t realized it yet.

Why it means "is dead"
Use ている after a verb such as 死ぬ to describe the resulting state. So 死んでいる means "is dead", not "is dying". The same grammar gives 結婚している, "is married".
Why "Nani?!" comes next
In the online meme, the victim responds with a shocked なにぃ!? ("Nani?!", "What?!"), just as Kenshiro’s opponents do in the series. The rough お前 and deadpan もう make the line sound final, which is what makes the joke work.
